Flash grenades

After playing through the game once, I read about two uses for the flash grenades that I wasn't aware of.

First: flash grenades kill the black birds that you see in a number of different areas, and causes them to leave behind money. Two excellent spots to use this: on the way to fight the first boss, there is a clearing (where you later fight a giant golem) and there is a circle of black birds on the ground. Throw a flash grenade to kill them, and you'll get a ton of money AND another flash grenade. The second spot is at a water fountain at the castle.

Second: I never knew this before, but flash grenades kill the Plagas (the alien creatures) instantly. This is especially handy at the farmhouse raid.

Raise Leons lifebar early in the game

This process requires you to use up several continues, which have no bearing on the game's ending. At the first El Gigante fight, there is a Yellow Herb in the boss arena. Take this item and combine it with any of the Green Herbs in the same room and use it. This increases Leon's lifebar slightly.

Now let the boss kill Leon. Since the boss' room is at a map transition (the green OPEN command is a dead giveaway), this lets you continue right at the boss fight but with Leon's life increase from your previous attempt. Repeating this process will increase Leon's lifebar to maximum prematurely. Extra Yellow Herbs found later can be given to Ashley, or sold to the merchant for money.

save ammo

to save ammo in the game when you have barely any ammo, shoot the zombies in the head and run up to them and press a to kick them or suplex them, which does massive damage, and if there is only 1, kick them down and cut them with your knife to save alot of ammo.

chainsaw maniac

when you first enter the village at the start of the game and you see the officer imppaled on the giant hook, in the fire, if you enter the house to the left, you will enter a cut scene and the villagers will start talking, and out will come a chainsaw weilding maniac that will cut your head off with a single swing and is extremely strong.

When you enter the village for the first time, you will find a shotgun upstairs in one of the houses. Get the shotgun and all the ammo you can find. You will obviously need to use your shotgun in the game, but try to save ammunition and use your pistol more. It may be harder, but the shotgun will definitely come in handy during large fights and boss fights.

To get the most items the first time around you need to find the most gold/money/gems you can to do this get the T. map from the merchant first its only 1000, you'll be able to buy your items faster this way
